Qualitative Analysis of Proteins Biuret Test Egg Albumin We require: egg albumin dispersion in a test
tube, sodium hydroxide solution, 1% copper sulphate solution,, test tube holder, rack,
droppers and a Bunsen burner. Take a small quantity sodium hydroxide solution
using a dropper. Add it to the egg albumin dispersion taken
in a test tube. Using another dropper, add a few drops of
1% copper sulphate solution to the test tube. Remove the test tube from the rack using the
test tube holder. Heat the contents of the test tube over the
burner for a few minutes. In the presence of alkali, proteins react
with copper (II) ions to form a violet colored complex called biuret. Gelatin We require gelatin dispersion in a test tube,
sodium hydroxide solution, 1% copper sulphate solution, test tube holder, rack, droppers

Take a few drops of Millon�s reagent using a dropper.
Add it to the egg albumin dispersion taken in a test tube.
A white precipitate is formed. Remove the test tube from the rack using a
test tube holder. Then boil the contents of the test tube over
a burner. White precipitate changes to brick red on
boiling, confirming the presence of proteins.
